What is being investigated

Directions the published work points at, and how far each has actually got. These are open questions, not established effects: most of what looks promising in cells does not replicate in animals, and most of what works in animals does not work in people. Nothing here is a claim about what this compound does for anyone.

  1. Whether cellular NAD+ can be raised at all

    The question everything else depends on, and the one with human data. Oral precursors produced dose-dependent increases in the blood NAD+ metabolome; the intact coenzyme is poorly taken up by cells, which is why the trials use precursors.

  2. Sirtuin- and PARP-dependent processes in ageing

    NAD+ is the required substrate for both enzyme families, and levels fall with age. Whether raising the substrate restores what the enzymes do is the actual hypothesis, and it is an animal-stage one.

  3. Neuroprotection and axon survival

    A separate strand, built on the finding that axon degeneration after injury runs through NAD+ depletion rather than alongside it. Studied in animal and cell models of nerve injury.

  4. Whether a measurable rise changes any outcome

    The gap between the first item and the second. Trials show the number can be moved; whether moving it does anything a person would notice has not been demonstrated in a controlled human trial.
