PT-141

A cyclic seven-amino-acid peptide, and an active metabolite of Melanotan-2 developed as a compound in its own right.

Also published as: Bremelanotide · Vyleesi

What is being investigated

Directions the published work points at, and how far each has actually got. These are open questions, not established effects: most of what looks promising in cells does not replicate in animals, and most of what works in animals does not work in people. Nothing here is a claim about what this compound does for anyone.

  1. Hypoactive sexual desire disorder in premenopausal women

The indication it is authorised in, on two randomised phase 3 trials across more than a thousand participants. What was authorised is a specific medicine, not this material.

    Human trials

  2. Erectile dysfunction

An earlier intranasal programme in men, discontinued after blood pressure rises were observed. That is the reason the compound went the direction it did, and it belongs beside the approval above.

    Human trials

  3. A central rather than vascular mechanism

What separates it from the PDE5 inhibitors: it acts at MC4R in the central nervous system rather than on vascular tissue, so the pathway is neural. Established in animal work.

  • PT-141 (bremelanotide): what the research says

Approved in the US as Vyleesi, with two phase 3 trials and a published safety programme. The mechanism is neural at MC4R, not vascular like PDE5 inhibitors.
